天螺霜理化性质及抗乙肝病毒作用的初步研究

     【摘要】 目的 探讨天螺霜的理化性质及抗乙肝病毒作用效果。方法 采用溶剂提取法自江西巴蜗牛提取天螺霜,复溶精制后作初步定性试验。采集32例HBV-DNA阳性患者血清标本各2份,分别加入天螺霜(试验组)和蒸馏水(对照组),37℃温箱中孵育3h后用PCR检测两组标本HBV-DNA含量。采用半体内法观察天螺霜对小鼠腹腔巨噬细胞吞噬功能的影响。 结果 天螺霜水溶液Pitrowsky试验阴性,碘液试验阳性,Molisch试验阳性。32例乙肝患者HBV-DNA体外实验观察结果显示,实验组、对照组阴转率分别为77.78%、0.00%,差异有显著性(P<0.01)。给药组和对照组吞噬百分率、吞噬指数分别为(61.7±4.73)%、0.89±0.071和(29.8±5.16)%、0.64±0.047,差异均有显著性(P<0.001)。结论 天螺霜为一类多糖物质,具有体外抑制乙型肝炎病毒复制的功能,可增强小白鼠腹腔巨噬细胞吞噬功能的作用。

    Preliminary study on physical and chemical characteristics and antihepatitis B virus function of Tianluopolysaccharides extract prepared from Jiangxinensis

    【Abstract】 Objective To study on physical and chemical characteristics and antihepatitis B virus function of Tianluopolysaccharides extract prepared from Jiangxinensis. Methods The organic element was prepared from Bradybaena.Jiangxinensis by solvent extraction methods and was qualitatively analyzed by Pitrowsky test, iodine test and Molisch test. Then we observed the inhibition effect of Tianluopolysaccharide on hepatitis B virus duplication in vitro with the method of polymerase chain reaction (PCR). The test of murine macrophages exposed to Tianlopolysaccharides was also carried out to obtain some insight into the interaction between Tianluo-polysaccharide and the functions of mouse peritoneal exudate macrophages. Results (1) The extract from Bradybaena.Jiangxinensis was some white and granulosum material that is soluble in water but not in alcohol. (2) The results of Pitrowsky test, iodine test and Molisch test were negative, brownish red color and purplish red color, respectively. (3) As the results of polymerase chain reaction (PCR) of hepatitis B virus (HBV) DNA, positive balteum appeared in every sample of control group and only 8 positive balteum appeared in testing group (P<0.01). (4) The index of phagocytosis(IP)and percentage of phagocytosis in testing group and control group were (61.7±4.73)%,(29.8±5.16)% and 0.89±0.071,0.64±0.047, respectively (P<0.001). ConclusionThe extract is initially identified as a polysaccharide material, and temporary named as Tianluopolysaccharide. In vitro test suggest that Tianluopolysaccharide exhibited antihepatitis B virus activity. The stimulation of phagocytic activity of macrophages was also proved by this study. ......
