CO2气腹对裸鼠腹腔种植肿瘤MMP-2和TIMP-1蛋白表达的影响(1)

     【摘要】 目的 评价CO2气腹及不同气腹压力对裸鼠腹腔种植肿瘤基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶抑制剂-1(TIMP-1)蛋白表达的影响。方法 30只裸鼠建立腹腔种植肿瘤模型,造模成功后随机分组:对照组、5 mm Hg和10 mm Hg CO2气腹组,气腹作用时间1 h。气腹干预后1周检测肿瘤组织MMP2、TIMP-1蛋白表达情况。结果 与对照组比较,气腹干预后1周5 mm Hg和10 mm Hg CO2气腹组肿瘤组织MMP-2蛋白表达均显著升高(P<0.01),且10 mm Hg组高于5 mm Hg组(P<0.01)。两组TIMP-1表达降低,且10 mm Hg组低于5 mm Hg组(P<0.01)。MMP-2/TIMP-1比例下降,10 mm Hg组下降更为显著(P<0.01)。结论:CO2气腹作用后早期可使肿瘤组织MMP-2升高,TIMP-1下降,气腹压力高作用更为明显。

    Effects of CO2 Pneumoperitoneum on Protein Expression of MMP2 and TIMP-1 of peritoneal implanted tumor in a nude mouse model

    【Abstract】 ObjectiveTo evaluate the effects of CO2 pneumoperitoneum and different pneumoperitoneum pressure on protein expression of MMP2 and TIMP-1 of peritoneal implanted tumor. MethodsThity nude mouse were divided randomly into three groups after the implanted tumor moel was established (control group, 5 mm Hg and 10 mm Hg CO2 pneumoperitoneum, 1 h). The correlated cytokines of aggression (MMP2/TIMP-1)in tumor tissue were detemined one week after pneumoperitoneum intervention. And to evaluate the effects of different CO2 pneumoperitoneum pressure on protein expression of MMP-2 and TIMP-1.ResultsCompared with the control group, the protein expression level of MMP-2 in the two CO2 pneumoperitoneum groups increased significantly in early post-operation(one week) (P<0.01). Moreover the expression level of MMP-2 in 10 mm Hg group was higher than that in 5 mm Hg group pneumoperitoneum(P<0.01). The protein expression of TIMP-1 decreased in both groups, with the lower expression in 10 mm Hg group(P<0.01). And the The expression level of MMP-2/TIMP-1 decreased after CO2 pneumoperitoneum intervention, especially in 10 mm Hg group(P<0.01).ConclusionThe expression of MMP-2 increase and TIMP-1 decrease in the early period after CO2 pneumoperitoneum intervention.
